Mutual Suppression of the Uricosuric Effects of Sulfinpyrazone and Salicylate: a Study in Interactions between Drugs.
نویسندگان
چکیده
A striking example of antagonistic drug effects (2) is afforded by the observation that in man salicylate, itself uricosuric in high dose, inhibits the pronounced uricosuria produced by probenecid (3, 4), sulfinpyrazone1 (5-9), and zoxazolamine (9-11). This suppressive action of salicylate has been assumed to be due to its successful competition with these drugs, and with uric acid, for renal tubular transport; one aspect of this competition is expressed as inhibition of tubular secretion of uric acid by small (and large) doses of salicylate, which partially counterbalances the inhibition of tubular reabsorption of uric acid produced by uricosuric agents (12). The present study provides experimental support for the operation of these mechanisms, but indicates that the interactions at least of salicylate and sulfinpyrazone are even more complex, involving competition also for binding sites on transporting plasma proteins, and possibly elsewhere. The consequences of the interplay of competition at these various sites are described in man, and also in the dog, in which mutual suppression of the uricosuric effects of sulfinpyrazone and salicylate, so apparent in man, could not be demonstrated.

Effects of uricosuric and antiuricosuric agents on urate transport in human brush-border membrane vesicles.
Inhibition of [14C]-urate uptake by uricosuric and antiuricosuric agents was investigated in human brush-border membrane vesicles, urate being transported either by anion exchange mechanisms or by voltage sensitive pathway. The IC50 for drugs on [14C]-urate uptake in vesicles loaded with 1 mM cold urate or with 5 mM lactate was, respectively: 0.7 and 0.3 microM for benzbromarone; 6 and 4 microM...

Pharmacologists believe analgesic and anti-inflammatory drugs such as non-steroidal anti-inflammatory drugs (NSAIDs) and opioid compounds, due to their side-effects and, in some cases, inadequacy, are not always useful. Therefore it seems necessary to search for newer analgesic compounds. In traditional Iranian Medicine, the leaf and rhizome of Sambucus ebulus is used as a topical medication ...
متن کاملPyrazinoate excretion in the chimpanzee. Relation to urate disposition and the actions of uricosuric drugs.
These experiments were designed to define the renal disposition of pyrazinoic acid in a nonhuman primate that is phylogenetically close to man and to relate this to the effects of pyrazinoate on urate excretion. The renal clearance of pyrazinoate was almost always greater than the simultaneous glomerular filtration rate at plasma concentrations ranging from 1.9 to 960 mug/ml. Some inhibitors of...

The effects of administration of drug combinations on uric acid excretion were studied in order to test the hypothesis that a portion of renal tubular reabsorption of uric acid occurs distal to the uric acid secretory site. Oral administration of pyrazinamide (3 g) during probenecid uricosuria (probenecid 500 mg every 6 h) decreased urate excretion from 463 mug/min following probenecid medicati...
